What studies are available related to the drug interaction between proton pump inhibitors (PPI) and clopidogrel (Plavix®) on cardiovascular (CV) related endpoints or outcomes?
This is part 2 of a 3 part series related to the drug interaction between proton pump inhibitors (PPI) and clopidogrel (Plavix®). Part 1 of this series described the functional activation of clopidogrel by the cytochrome P450 (CYP) enzymes as well as the differences that PPI can have on those enzymes.(1) The last two issues in this series will focus on the translation of the pharmacokinetic data presented in part 1 to patient-oriented related outcomes seen in available studies.
A number of studies have now been reported linking concomitant PPI and clopidogrel use with worse cardiovascular (CV) related endpoints or outcomes which include rehospitalization for acute myocardial infarction (AMI), acute coronary syndrome (ACS), need for revascularization and even death (see table below).(2-6) However, much of this data come from....
